What is more sound dampening, 1 layer of drywall or 1 layer of mass loaded vinyl?

Don’t want to open too much of a discussion (I am well aware of sound dampening limitations), I only have few options to work with unfortunately.

I am going to fill the studs in an open ceiling with either:

1 layer of dry wall + insulation.

Or

1 layer of either 1lb or 1/2lb mass loaded vinyl + insulation.

Which will reduce the most noise from upstairs?

Mass is what stops sound. Always go with whatever has more mass. If you dont create a decoupled MSM system, then you are at the mercy of the mass law. Every doubling of mass adds 6db of isolation (that's not good).

Hmmm okay, so I guess with a (very) non professional installation the dry wall is the way to go. Thank you.

You're welcome. You could possibly get better/more complete advice, if you provided more details on your situation.

Finishing off a basement room and I am just trying to reduce a little noise from upstairs and vice versa. No chance of doing any complex soundproofing and can’t close the ceiling off.

I’m thinking I may just stuff rock wool between the studs, maybe it is just more practical.

Gotcha, yeah, if you cant close it off, theres not much you can do to increase isolation substantially. I would caulk all seams and add some semi rigid insulation batts between the joists.

What drywall? 5/8" drywall weighs about 1 lb/ft3. It costs about 65 cents per ft2. MLV about 7 dollars/ft2.
