I'm a totally OTB kind of guy simply because I think music is kind of an organic process like cooking. So, I seriously doubt I'm going to get the same musical experience by staring and tweeking layers in an audio application. I don't even like keyboards.
However, non-linear editing came about for a reason. This, like Izotope RX, is another amazing utility that would be really foolish to ignore as useless. The scary thing is that odds are people would use this solely as a source for creation without any kind of live creation at all. I think that sounds like fun too, but I doubt I'd ever get around to doing things that way (See first paragraph).
I think people take things to extremes and forget the basis of natural ingredients that make a sound what it is, just like seeing a live show versus an mp3 on myspace. The expense of recording sound naturally also helps push that notion further from existence.
Overall, I think Melodyne's Direct Note Access is simply amazing. It's an instrument of it's own.
